Beating out Joss Stone, Jamelia, Corinne Bailey Rae and Beverley Knight with her Back to Black album, Winehouse also took to the stage to a standing ovation before performing "Me and Mr Jones" and "Tears Dry On Their Own".
Winehouse's award victory at the Mobos - Music of Black Origin Awards - comes after months of reported problems with alcohol and drugs, prompting her to check into a rehab clinic with her husband Blake Fielder-Civil last month.
The 24-year-old also pulled out of concerts in Britain and France because of "health issues" and was admitted to hospital last month over what her record company said was severe exhaustion.
Meanwhile, Dizzee Rascal was named Best UK Male for his Maths and English album, and Kanye West went home with two awards - Best Video for "Stronger" and Best Hip-Hop.
Barbados-born teenage sensation Rihanna, who earlier this year topped the British singles charts for 10 weeks running with "Umbrella", was named Best International Act.
