The Victorian had a fantastic start before running away from the field at the 60 metre mark to finish a clear two metres in front of his nearest competitor posting a time of 12.13 seconds.
The 21-year-old, competing in his fourth Stawell Gift, held off main rival Richard Hankin from New South Wales with Ben Vickery third and national 100 metres champion Otis Gowa fourth.
Jamieson said it was the biggest achievement in his athletics career.
"I'm over the moon at the moment, it's all surreal, and it's probably going to take me quite a bit of time to take it all in," he told channel 10.
"I ran well in the semi, felt good coming into the final and knew i just had to hit the gun - I've done it in training, I just had do it on the day."
